If you have a hose or part of the gauge which takes a lot of air - yes. I learnt this on my mountain bike air shocks which need fine adjustment - inflate the shock (or tyre) to the pressure you need (I have a combined pump and gauge), the pressure will be constant across the volume of the shock (or tyre in your case) and internals of the gauge. When you reconnect the gauge an amount of air will enter the pump so the pressure will go down (as long as nothing else has changed e.g. temperature), the decrease in pressure depending on the volume of air required by the gauge.
