If the tyre's bead is not making enough contact with the rim to enable an initial seal, remove the tyre again and pack the gap between each bead with something like folded newspapers so that you are streching the tyre walls outwards.Place the tyre in a very warm place and allow the tyre to expand to the stretching. Allow it to cool. When you refit the tyre it will seal when you inflate it. Remove the valve to allow a greater inflation force. The tyre has probably been standing with something crushing the sidewalls.
