Usually measured by placing a piece of solder wire through the plug hole and bent 90 degrees to reach the side of the cylinder bore with the piston somewhere in the lower part of the barrel. The engine is then turned over to crush (or maybe Squish!) the solder allowing the gap between the piston and head to be measured.

There are usually minimum measurements for each class as the smaller the gap the higher the compression ration.
