To answer your initial question, if your axle is straight and the sprockets are running true with no excessive or un-even sprocket wear.......then its due to the un-even wear in the chain links, this is quite a common problem.

When your setting your chain tension keep rotating the wheel and set the chain tension at the tightest spot to about 15mm total play......the rest of the chain-play in the slacker regions should be about 25mm, hope that helps?
