First thing I would do is pressure test the carb to see if it is leaking. If you have access to another carb it would be good to test to see if it solves the problem. At least you can then rule out the carb. I would then check the reeds for holes, damage or not meeting correctly. Are you using standard reeds or carbon ? The carbon ones will give better throttle response but are prone to getting damaged especially if you get a backfire. Next I'd check the piston ring to see if it's stuck and needs freeing off.
I assume you're asking because you've checked the very obvious things like gearing, brake binding, carb rebuilt correctly and not ready for a top end rebuild yet.
